Salt Lake City, March 1, 2005 -Myriad Genetics, Inc. (Nasdaq: MYGN), announced
today that it will begin a second Phase I clinical trial with its investigative cancer
drug, MPC-6827, under an FDA-approved Investigational New Drug application.
This new human clinical study will evaluate the potential of MPC-6827 to treat
metastatic brain cancer by achieving therapeutic concentrations in the brain that are
sufficient to treat tumors without significant systemic exposure or toxicity. The

Structure and function of the brain
The brain , together with the spinal cord, is the control centre which governs how the body functions. The basic cell of the brain is the nerve cell (neurone); you are born with the total number of nerve cells and, unlike other cells, they do not replace themselves as they die. The nerve cells communicate with each other and other parts of the body by sending messages along the nerve pathways or by controlling the release of hormones.

Primary brain tumors can either be benign or malignant
Malignant brain tumors, such as glioblastomas tend to grow rapidly spreading into the surrounding brain tissue with the help of proliferating core and invasive rim cells which cannot be entirely removed surgically.

of the incident (newly diagnosed) cancers reported to the Ohio Cancer Incidence

tumor cells. It's a new class of drugs called "immunotoxins" -- consisting of a bacterial toxin that has been genetically
altered and combined with a protein that makes the killer bacteria avoid normal cells and attack only the cancerous cells in
the brain .

DURHAM, N.C. -- In a daring yet successful experiment to cure deadly brain tumors, researchers have combined the
cancer -killing properties of poliovirus together with a harmless genetic coding element from the common cold.
The resulting modified virus created a remarkably strong anti- cancer agent that rapidly killed cancer cells in laboratory cell
cultures and in animals -- and without causing polio, said Matthias Gromeier, M.D., assistant professor of molecular
genetics and microbiology at the Duke Comprehensive Cancer Center. Testing of the new viral agent in humans should
begin within two years, he said.
In the study, the modified poliovirus rapidly killed cancer cells derived from primary brain tumors as well as cells derived

• Many of the cases initially suggested by the union were ruled out because they were not malignant brain
tumors. Some were brain tumors that were either benign (non-cancerous) or were a metastasis from an-
other type of tumor. The brain is a common site to which a malignant tumor (a metastasis), which be-
gins in another part in the body, can spread. Only malignant brain tumors were considered part of the
investigation. Benign brain tumors have different risk factors.
• There were four cases of primary, malignant (cancerous) brain tumors confirmed among white male
employees at the P&W North Haven facility during the period 1991-1999. Three of these were the

The machinists union contacted the Connecticut Department of
Public Health to assist them in evaluating a concern of their mem-
bers and families regarding a cluster of brain cancer cases at the
North Haven facility. Those who were concerned had developed
a list of persons they suspected had brain cancer who had worked
at the facility. Since the number of cases was consistent with a
need for follow-up, the DPH conducted a formal investigation
during the summer and fall of 2000.
There were several purposes in conducting the study:
• Confirm the diagnoses of these cases medically. If tumors
were found, see if they were malignant brain tumors.
• Determine if there were any other cases.
• Determine whether there were more cases of brain cancer oc-
curring in P&W employees than would be expected based on
comparison with the overall population in Connecticut. ...

New USC neuro-oncologist seeks
new ways to fight brain cancer
The story seems apocryphal, she ad-
mits.
When Kathleen Squires was a little
girl growing up overseas in places like
Indonesia, she saw people around her
with elephantiasis—enlarged, bulging
legs due to lymphatic infection—and
asked her dad why they suffered from
the deforming condition.
“They don’t have good medical
care,” he explained.
And so, though no one in her family
worked in medicine, she decided she
would be a doctor.
Today, she is one of the newest medi-
cal faculty members of the Keck School
of Medicine of USC, focusing on infec-
tious diseases. Specifically, Squires—
visiting associate professor of medi-
cine—treats patients with HIV and
AIDS and performs research on drugs
that might better help such patients.

#266 - Brain Cancer Is Rapidly Increasing

Brain cancer is relatively rare, accounting for only 2% of all cancers,
but it seems to be increasing rapidly in children and in people over
65 in the U.S. The latest figures from the National Cancer Institute
indicate that brain cancer in the elderly (ages 75-84) is doubling
every 9 years [1, pg. 1621; 2, pg. 635.] In 1991 in the U.S., an
estimated 16,700 new cases of brain cancer were diagnosed and an
estimated 11,500 deaths occurred. [3, pgs. I.15-I.16] The causes of
brain cancer are unknown.
Brain cancer is usually fatal; the average (median) time between
diagnosis and death is about nine months. [1, pg. 1621.] The average
survival rate after 5 years is 25%, but in people over 65 the 5-year
survival rate is only 4%. [3, pg. I.16.] ...
