OCTOBER 24, 2005 Blood Test Shows Promise in Early Identification of Colon Cancer Researchers have developed a blood test that detects the presence of fragments of mutated genes that are present in colon cancer cells. Their pilot studies provide the basis for diagnostic assays for cancer that could allow physicians to detect tumors at an early stage. The research team, led by Howard Hughes Medical Institute investigator Bert Vogelstein at The Sidney Kimmel Cancer Center at the Johns Hopkins Medical Institutions, published its findings on October 24, 2005, in the online Early Edition of the Proceedings of the National Academy of Sciences (PNAS) . Researchers have known for years that abnormal activity of tyrosine kinases can hasten the development of certain forms of cancer . In this first-of-a-kind survey, researchers used a forward-looking approach to screen the genes that produce tyrosine kinases — reasoning that somewhere in this genomic landscape they might encounter novel mutations that spur colon cancer and other types of cancer . Telomerase activation, a cardinal requirement for immortalization, is a crucial step in the development of malignancy and requires the induction of the catalytic component, human telomerase reverse transcriptase (hTERT), encoded by the hTERT gene. By reverse transcription-PCR, using primers within the reverse transcriptase domain of hTERT, we investigated telomerase messenger in 8 adenomatous and 9 dysplastic polyps, and in 32 paired cancer -normal mucosa specimens, one liver and one spleen metastasis from patients resected for sporadic colorectal cancer . Telomerase messenger was absent or very low in normal mucosa and in adenomatous polyps. Dysplastic polyps and adenocarcinoma samples showed hTERT mRNA, with higher levels in cancer tissues compared to dysplastic lesions. The incidence of colorectal cancer in Hampton Roads is on the increase. In fact, the death rate for colon can- cer in most cities in Hampton Roads exceeds state and national rates. The ACS recommends colorectal cancer screening for men and women starting at age 50. Although the exact cause of colorectal cancer is unclear, experts believe most cases begin as polyps--small, precancerous growths on the inner wall of the colon or rectum.
