Background: New tumor markers and markers of tumor
progression are needed for improved staging and for better
assessment of treatment of many cancers. Gene expression
profiling techniques offer the opportunity to discover such
markers. We investigated the feasibility of sample pooling
strategy in combination with a novel analysis algorithm to
identify markers. Methods: Total RNA from human colon
tumors (n = 60) of multiple stages (adenomas; cancers with
modified Astler Collier stages B, C, and D; and liver metas-
tases) were pooled within stages and compared with pooled

Targeting colon cancer cells with genistein-17.1A immunoconjugate

Abstract.
We have shown that genistein, a major component
of soy, has anti- colon cancer effects
in vitro . These effects are
attainable at high concentrations that are difficult to achieve
in the serum. The purpose of this study was to enhance the
activity of genistein against colon cancer cells by coupling it
to 17.1A. The monoclonal antibody 17.1A recognizes an
epithelial membrane antigen that is overexpressed in colon
cancer . Synthesis of Gen-17.1A was achieved by photo-
chemical conjugation using sulfa-SANPAH. Its purity was
evaluated by SDS-PAGE. Binding of Gen-17.1A to SW-
620 and HT-29 cells was shown using flow cytometry.
Internalization was demonstrated by FITC-labeling. Gen-
17.1A induced apoptosis in colon cancer cells as evidenced
by the acridine orange/ethidium bromide staining method.

Abstract.
The chemoprevention of colon cancer , the second
leading cause of cancer -related deaths in the United States,
has been pursued actively during the last two decades.
Methodological problems and those stemming from the
available agents per se have constrained this difficult task;
to some extent, these problems persist to date. That
NSAIDs decrease the incidence of and mortality from colon
cancer has been a major advance in chemoprevention. These
compounds are, however, limited by their significant side
effects. NO-releasing NSAIDs (NO-NSAIDs) are a novel
class of compounds, synthesized to overcome the limitations
of NSAIDs. In general, they appear safer and much more
effective than their traditional counterparts. We review their
structural features, metabolism and pharmacological actions.

Abstract.
MSH2 and MLH1 are proteins involved in DNA
reparation. They are mutated in some forms of colon cancer ,
i.e. hereditary non-polyposis carcinomas and a subset of
sporadic carcinomas. We have studied the expression of
MSH2 and MLH1 in a retrospective series of 225 colorectal
carcinomas by immunohistochemistry. The results were
compared to molecular tests of microsatellite instability using
amplification by PCR of BAT-25 and BAT-26 repeated
sequences and to histoclinical data. Positivity of both proteins
was never associated with MSI phenotype. MSH2 and/or
MLH1 negative tumors were frequently tumors of the proximal
colon ; in this subpopulation or proximal tumors, MSH2
and/or MLH1 negativity was associated with a longer disease-
free survival.

Abstract.
The current chemotherapeutic modalities for
advanced colorectal cancer are limited. DNA-platinating
drugs such as cisplatin have poor efficacy against this
malignancy. The aim of this study was to identify genes
that render survival advantage after cisplatin treatment in
metastatic colon cancer . Cell lines SW480 (primary colon
cancer ) and SW620 (metastatic lesion from the same patient)
were obtained from ATCC. Apoptosis was measured by FACS
analysis of cisplatin-treated (0.01-10
µ g/ml) and untreated
cells. Simultaneous analysis of ~1200 cDNAs was performed
by microarray technique on untreated and treated cells from
lines. Microarray results were confirmed by RT-PCR. The
SW620 cell line was more resistant to apoptosis induced by

Abstract.
Telomerase activation, a cardinal requirement
for immortalization, is a crucial step in the development of
malignancy and requires the induction of the catalytic
component, human telomerase reverse transcriptase (hTERT),
encoded by the hTERT gene. By reverse transcription-PCR,
using primers within the reverse transcriptase domain of
hTERT, we investigated telomerase messenger in 8
adenomatous and 9 dysplastic polyps, and in 32 paired
cancer -normal mucosa specimens, one liver and one spleen
metastasis from patients resected for sporadic colorectal
cancer . Telomerase messenger was absent or very low in
normal mucosa and in adenomatous polyps. Dysplastic
polyps and adenocarcinoma samples showed hTERT mRNA,
with higher levels in cancer tissues compared to dysplastic

Cancer of the colon and rectum can be
deadly. It can strike anyone. But the good
news is that colorectal cancer can be
prevented. The key is to find and remove
tumors before they turn cancerous.
Beginning at age 50, testing for colorectal
cancer includes one of the following:
I Sigmoidoscopy—every five years.
I Colonoscopy—every 10 years.
I Double-contrast barium enema—every
five to 10 years.
Each year your doctor should also give
you a test to do at home that checks for
blood in the bowel movement. This is
called the fecal occult blood test, or stool
test.
These tests are the most current
screening guidelines of the American
Cancer Society and are recommended for
members of Santa Barbara Regional Health
Authority.

PURPOSE: The purpose of this research was to investigate the frequency of colorectal cancer
screening African-American women aged 50 and older who are in the high risk category for
developing colorectal cancer and polyps. Additionally, their beliefs and attitudes were examined
through the guidance of the Health Belief Model to aid in the prediction of screening utilization.
METHODOLOGY: This randomized study used a descriptive/correlative, non-experimental

DEFINITION
The colon (large intestine) and rectum make up the final segment of the digestive tract. It is the lower four to
five feet of the intestine that travels around the borders of your abdominal area. The rectum is the last four to
five inches at the end of the colon . Cancer can develop anywhere along the digestive tract. Cancer of the colon
and rectum is called colorectal cancer . Colorectal cancer is potentially one of the most curable cancers if detected
early.
