Acute Myeloid Leukaemia (AML)
What is leukaemia ?
Leukaemia is a cancer of cells in the bone marrow (the cells which develop into blood cells).
Cancer is a disease of the cells in the body. There are many types of cancer which arise from
different types of cell. What all cancers have in common is that the cancer cells are abnormal and
do not respond to normal control mechanisms. Large numbers of cancer cells build up because
they multiply 'out of control', or because they live much longer than normal cells, or both.
With leukaemia , the cancerous cells in the bone marrow spill out into the bloodstream. There are
several types of leukaemia . Most types arise from cells which normally develop into white blood
cells. (The word leukaemia comes from a greek work which means 'white blood'.) If you develop
leukaemia it is important to know exactly what type it is. This is because the outlook (prognosis) ...

A one-day meeting organised by the RMS and the Institute of Cancer Research
The Conference Centre, Royal Marsden Hospital, London
Monday 26 April 2004
Organiser: Ricardo Morilla, Department of Haemato-Oncology, Royal Marsden Hospital
This well established annual flow cytometry meeting on immunophenotyping is ideal
for people in the field who like to remain updated on the recent advances in
immunophenotyping and flow cytometry in general.
The aim of the meeting is to discuss the main aspects of the use of flow cytometry in
the diagnosis of leukaemia and lymphoma. The intention is to present a balanced
program which will include both research developments in the field and routine
problems encountered by those using flow cytometers on a daily basis. The meeting
will be of interest to experienced individuals and to those ...

HIV-1 INFECTION IN A PATIENT OF ACUTE LYMPHOCYTIC
LEUKAEMIA MISSED BY HIV-SPOT AND HIV-SCAN RAPID TESTS
*S Singh, V Kumari, N Singh
Abstract
A case of HIV-1 infection, presumably acquired through unsafe blood transfusion in an acute
lymphocytic leukemia (ALL) patient, is reported here in whom the two rapid HIV antibody detection
kits (HIV-spot and HIV-Scan) failed to diagnose the infection while the sample was strongly positive
in microwell ELISA (detect- HIV) on the same day and was confirmed by western blot assay (NEW
LAV BLOT-1). However, repeat sample after 15 days was found positive in all the tests. Though the
rapid tests are preferred over microwell ELISA because of ease of performance and rapid availability
of results, this report confirms low sensitivity of the two rapid tests. This suboptimal sensitivity,
particularly in cases of early seroconversion, therefore, must ...

Abstract. More than a decade of experimental work in
an inbred subline of Sprague-Dawley rats having high
incidence of spontaneous T-cell lymphoma/ leukaemia is
reviewed. Longitudinal follow-up of biological charac-
teristics (growth, survival, haematology) of both multi-
ple cases of primary disease and s.c. passaged lym-
phomas as well as comparative immunophenotypic and
karyotypic studies are concluded. In these T-cell lym-
phomas (mostly CD4 positive), arising on the same
genetic background of the inbred SD strain, the aberra-
tions involving chromosome 11 have been recognized as
a typical non-random cytogenetic marker. This unique
rat model of lymphoblastic lymphomas/leukaemias, rel-
evant to human pathology, seems to be very suitable for
testing different anticancer therapeutic strategies, as it is
documented by results of a number of various protocols
conducted ...

Abstract
The evidence linking indoor radon exposure to childhood and adult leukaemia is reviewed. At the UK
average indoor exposure of 20 Bq m
-3
, it is estimated that the radon derived equivalent dose accrued to the
fetus is 106 µSv. The equivalent dose rate to adult bone marrow 130 µSv y
-1
. Standard radiation risk
factors suggest that 5% of childhood and 4% of adult leukaemia is linked to radon at 20 Bq m
-3
exposure.
Geographical studies generally support such a link at about this magnitude. A number of case-control studies
have been carried out but ...

Abstract
Acral erythema or erythrodysesthesia is a common occurrence in patients of AML treated with high dose cytarabine where bullae
formation may be a part of its evolution.Though, most of the time it is a self limiting condition, rarely it may be severeenough to
cause ulceration of the digits and may give rise toseveredistress to the patient possibly due to ischaemia.Avoidance of the culprit
drug is required in such a severesituation.

Abstract.
Spontaneous remission of haematologic malignancy is extremely rare. Spontaneous remission
of chronic lymphocytic leukaemia (CLL) has been associated with viral infection, immunization and new
primary carcinoma. We report two cases of spontaneous regression of CLL, observed after 20 and 9
years from diagnosis. Spontaneous regression of CLL was documented by clinical and haematological
data and was not associated with viral infection or secondary malignancy. Both patients are alive and
without signs of CLL, three and four years after spontaneous regression of the disease. Authors discussed ...

Introduction
This presentation is a case-history of an attempt by a local health authority in the U.K. to deal
with high levels of public anxiety about a cluster of childhood leukaemia in Northampton by a risk
communication strategy. Usually, in similar circumstances of intense public demand and extensive
adverse publicity, health authority’s have responded by setting up, predictably inconclusive,
epidemiological studies. Despite intense public, political, media and management pressure to do
just this, we chose rather to educate the public about disease clusters and the limitations of
epidemiological methods. This involved working with the community ...

Abstract. In order to assess the effect of the DNA polymerase
inhibitor aphidicolin on resistance to cytosine arabinoside,
blast cells from 15 children with ALL and 9 with AML were
exposed to a range of concentrations of ara-C ± aphidicolin.
Cell survival was measured using the MTT assay. Aphidicolin
significantly increased sensitivity to ara-C in blast cells from
both ALL (p=0.001) and AML (p<0.01). The median fold
increase (sensitisation ratio) for ALL was 3.4 (range 1.2-13.6)
compared to 12.4-fold (range 6.0-148) for AML blasts
(p=0.005). There was a striking relationship between increasing
ara-C resistance and increasing effect of aphidicolin in AML
(p<0.001) but not ALL (p>0.05). These remarkable results
suggest that aphidicolin should be considered for future
clinical trials as a modulator of ara-C resistance, particularly
in AML.
