44 P&T ® • January 2003 • Vol. 28 No. 1 A one-day workshop, entitled “Innovations in Lymphoma Treatments,” was held on Novem- ber 16, 2002, in New York City. The sessions provided a forum for patients, caregivers, and clinicians to learn about current and future clinical tri- als in lymphoma research. The workshop also provided an opportunity to gain knowledge of the promising new therapies under clinical investigation and to reflect on the combination of currently approved therapies with new drug classes to treat follicular B-cell non-Hodgkin’s lymphoma (NHL) and other lymphomas. This is an era of exciting new de- velopments in drugs and biologicals for the therapy of NHL; breakthroughs will eventually prolong the lives of patients of all ages with the disease. A new age of lymphoma therapy began in 1997 with the ap- proval of rituximab ...

Nuclear Medicine: Third of Lymphoma Patients in Remission with Bexxar - a smart isotope More than a third of lymphoma patients who had previously failed other therapy have no detectable signs of the disease three years after being treated with the experimental drug Bexxar, researchers said on Monday. Bexxar combines a natural immune system targeting agent with radiation therapy in the form of iodine 131, a radioactive isotope. The combination allows the tumor cells to receive a greater dose of radiation, while minimizing the amount of radiation affecting normal tissues, including delicate bone marrow. The drug is being developed by Coulter and SmithKline Beecham, which filed in September for U.S. Food and Drug Administration approval of the product. Research presented at a meeting of the American Society of Hematology in San Francisco showed that of 251 low-grade non-Hodgkin's lymphoma patients injected ...

There is possible evidence from studies conducted at Los Alamos National Laboratory. Studies of nuclear workers at other sites who have been exposed to ionizing radiation and persons exposed to the atomic bomb show an increase risk of developing non-Hodgkin's lymphoma . The National Research Council’s BEIR V committee did not address the issue of radiation- induced non-Hodgkin’s lymphoma . Non-Hodgkin's lymphoma is designated as a “specified” ...

7 BACKGROUND INFORMATION ON NON-HODGKIN’S LYMPHOMA General Non-Hodgkin’s lymphomas (NHLs) encompass several unique malignant lymphoid disease entities that vary in clinical behavior, morphologic appearance, immunologic, and molecular phenotype. The various types represent neoplastic lymphoid cells arrested at different stages of normal differentiation. Based on their natural history, NHLs can be clinically classified as indolent, aggressive, and highly aggressive. Epidemiology NHLs are the fifth most common cause of cancer in the United States, with an estimated incidence of 63,600 cases in 2001 i . Follicular center cell lymphomas are the second most common subtype, comprising approximately 40% of all non-Hodgkin’s lymphomas. Since 1950, the incidence of NHL has steadily increased at approximately 4% per year. Classification Several histologic classifications ...

Persistent SPN early 2001 with no other evidence of disease. Why PET was ordered : This is the patient’s 1 st PET scan. It was ordered to evaluate SPN to recurrent lymphoma vs. new malignancy vs. benign change. PET Findings: SPN with increased uptake. Otherwise negative. Patient Treatment Management Change: Positive SPN, with no evidence of lymphoma elsewhere, justified surgery . Surgery proved that the nodule was in fact early stage low-grade lymphoma . This was different from the original histology which could be completely resected. Patient received their 3rd PET scan 8 months later: Following CT scan ...

Hematology 1999 329 Mantle Cell Lymphoma : New Treatment Possibilities Bertrand Coiffier Mantle cell lymphoma (MCL) represents one of the lym- phomas most refractory to treatment. Its initial presen- tation is characterized by a slow increase of lymph node volume, spleen size, and blood involvement making it a real indolent lymphoma . At diagnosis, most of the pa- tients have disseminated disease with enlarged lymph nodes, bone marrow and blood involvement, splenom- egaly, and often involvement of extranodal sites such as the colon or Waldeyer’s ring. The incidence of this lym- phoma is similar in the US and Europe, representing 8% of the non-Hodgkin’s lymphomas. Very few prospective clinical trials exist, making it impossible to define rec- ommendations for the treatment of these patients. More- over, the interpretation of the clinical trials or retrospec- ...

With contemporary imaging, staging, and treatment, primary lymphoma of bone is thought to have a good prognosis. We performed a retrospective analysis of patients who had lymphoma of bone to confirm or deny this impression, to determine with contemporary staging how many patients had stage IE, primary lymphoma of bone versus advanced lymphomatous disease, and to evaluate the prognostic factors and oncologic outcome of adult patients who have lymphoma of bone. Non-Hodgkin’s Lymphoma 70 CANCER INCIDENCE AND MORTALITY IN NEW MEXICO, 1970-1996 Major Epidemiologic Features Lymphomas are cancers of the white blood cells (lymphocytes) of the immune system. The disease process involves an abnormal growth of lymphocytes either within the lymph nodes or at extra-nodal sites such as the spleen and thymus. Non-Hodgkin’s lymphoma (NHL) refers to a diverse group of lymphomas that include many different histologic types, but exclude the cell types associated with Hodgkin’s disease, another form of lymphoma . NHL is a relatively common cancer, accounting for 4 to 5% of all cancers diagnosed in the U.S. each year. The incidence of NHL has increased dramatically over the past several decades in the U.S., roughly doubling between 1973 and 1995. Part of the increase in recent years has been related to the acquired immunodeficiency disorder (AIDS) epidemic. NHL is about 60 ...

Protection from Lymphoma Cell Metastasis in ICAM-1 Mutant Mice: A Posthoming Event Fawzi Aoudjit, 1 * Edouard F. Potworowski,* Timothy A. Springer, † and Yves St-Pierre 2 * It has been hypothesized that the intercellular adhesion receptors used by normal cells could also be operative in the spreading of circulating malignant cells to target organs. In the present work, we show that genetic ablation of the ICAM-1 gene confers resistance to T cell lymphoma metastasis. Following i.v. inoculation of LFA-1-expressing malignant T lymphoma cells, we found that ICAM-1-deficient mice were almost completely resistant to the development of lymphoid malignancy compared with wild-type control mice that developed lymphoid tumors in the kidneys, spleen, and liver. Histologic examinations confirmed that ICAM- 1-deficient mice, in contrast to wild-type ...

From the GRCA Health and Genetics Committee GOLDENS WITH LYMPHOMA AND OSTEOSARCOMA ARE NEEDED TO STUDY THE GENETIC BASIS OF LYMPHOMA AND OSTEOSARCOMA Most Golden Retriever fanciers know Goldens that have died of lymphoma (cancer of the lymph nodes, lymphosarcoma) and osteosarcoma (bone cancer). Since our Goldens are predisposed to these cancers, many of us have lost some of our own beloved Goldens to these diseases. The Canine Health Foundation has funded a study that may help determine the genetic basis of these cancers. We have hopes that this study and subsequent studies may some day lead to genetic tests to identify Goldens predisposed to developing lymphoma and bone cancer and may identify those lymphomas and bone cancers most likely to respond to treatment. If your Golden Retriever is newly diagnosed with lymphoma or osteosarcoma (has not yet started chemotherapy, including steroids), ...

MOLECULAR ASSESSMENT OF GENE EXPRESSION IN B-CELL LYMPHOMA OF SIV INFECTED RHESUS MONKEYS ( MACACA MULATTA ). AN INTRODUC- TION. BODEMER, W., FLOTO, A., MÄTZ-RENSING, K. AND KAUP, F.-J. Key words: B-cell lymphoma , Simian immunodeficiency virus, HIV, oncogenic her- pesvirus, differential gene expression Abstract B-cell Non-Hodgkin’s lymphoma (B-NHL) is a typical tumor after infection with viruses causing immunodeficiency. Both, Simian as well as Human Immundeficien- cy Virus (SIV and HIV) are well known pathogens suspected to contribute to the de- velopment of B-cell lymphoma . Fortunately, macaques infected with SIV were shown to be an adequate non human primate model for NHL. In our group we established profound pathological examination protocols and identified classification markers. We conducted experiments to further insight into the molecular processes of lympho- magenesis. Presence of simian ...

68 NATURE MEDICINE • VOLUME 8 • NUMBER 1 • JANUARY 2002 A RT I C L E S Diffuse large B-cell lymphomas (DLBCLs) are the most common lymphoid neoplasms, composing 30–40% of adult non-Hodgkin lymphomas 1 . Although a subset of DLBCL patients are cured with current chemotherapeutic regimens, most succumb to the disease 2 . Clinical prognostic models such as the International Prognostic Index (IPI) have been developed to identify DLBCL patients who are unlikely to be cured with standard therapy 3 . However, the clinical factors of the IPI (age, performance status, stage, number of extranodal sites and serum lactate dehydroge- nase (LDH)) 3 are likely to be surrogate markers for the intrinsic molecular heterogeneity in this disease. Therefore, it is not sur- prising that IPI is imperfect in its identification of high-risk pa- ...
