Copyright © 1998 by W.B. Saunders Company. All rights reserved. 320 A BASIC INFORMATION DESCRIPTION A malignancy beginning in the plasma cells of the bone marrow. Plasma cells normally produce antibodies to help destroy germs and protect against infection. With myeloma , this function becomes impaired, and the body cannot deal effectively with infection. It can affect the bone marrow of all bones but is most common in the thigh, back, pelvis or upper arms. It is most com- mon in men between ages 50 and 70. A recent report has found a virus associated with multiple myeloma . FREQUENT SIGNS AND SYMPTOMS • Pain in the affected bone. The pain is severe, boring and deep. If the bone collapses, pain spreads to other parts of the body. • Weight loss. • Symptoms of anemia, such as weakness, paleness, tiredness and breathlessness. CAUSES Unknown. ...

Multiple Myeloma 66 CANCER INCIDENCE AND MORTALITY IN NEW MEXICO, 1970-1996 Major Epidemiologic Features Multiple myeloma is a rare cancer that arises in the bone marrow and involves the plasma cells, a type of blood cell. Plasma cells play a critical role in the immune system by producing antibodies which normally circulate in the blood to ward off disease. The main characteristic of multiple myeloma is an overgrowth of malignant plasma cells that leads to the excess production of a specific antibody or component of the antibody. Multiple myeloma accounts for about 1% of all cancers diagnosed each year in the U.S., and about 2% of all annual cancer deaths. The prognosis for multiple myeloma is generally poor, with the probability of dying within five years of diagnosis at about 70%. Over the past several decades in the U.S., incidence has been rather stable while mortality has been slowly ...

Pathophysiology of Multiple Myeloma (MM) •Many problems associated with MM include: --plasma cell tumor that secrete substances; --synthesis of large quantities of monoclonal antibodies. •Most common presenting features in MM are: --bone pain; --hypercalcemia; --osteolysis; ...

MULTIPLE MYELOMA Pennsylvania Department of Health - Pennsylvania Cancer Incidence and Mortality 1994-1998 - Page 196 Incidence rates for multiple myeloma reported from the National Cancer Institute's SEER Program show the highest risks in blacks. The 1994-98 average annual age-adjusted incidence rates for Pennsylvania showed the same relationship. The rates for black male and black female residents were 11.4 and 7.9, respectively, versus 4.3 per 100,000 for all reported cases. Multiple myeloma accounted for 769 cancer cases diagnosed among Pennsylvania residents in 1998, the highest annual number reported during to ten-year period of 1989-1998. Annual age-adjusted incidence rates between 1989 and 1998 have generally been on the increase among all four sex/race groups. A distinct feature of multiple myeloma is the strong association with age. The average annual (1994-98) age-specific incidence rates were ...

NEOPLASIA The myeloma -associated oncogene fibroblast growth factor receptor 3 is transforming in hematopoietic cells Zhihua Li, Yuan Xiao Zhu, Elizabeth E. Plowright, P. Leif Bergsagel, Marta Chesi, Bruce Patterson, Teresa S. Hawley, Robert G. Hawley, and A. Keith Stewart Translocations involving fibroblast growth factor receptor 3 (fgfr3) have been identi- fied in about 25% of patients with my- eloma. To directly examine the oncogenic potential of fgfr3, murine bone marrow (BM) cells were transduced with retroviral vectors containing either wild-type fgfr3 or an activated mutant form of the recep- tor, fgfr3-TD. Mice transplanted with FGFR3-TD–expressing BM developed a marked leukocytosis and lethal hemato- poietic cell infiltration of multiple tissues within 6 weeks of transplantation. Second- ary and tertiary recipients of spleen or BM from ...
