Proteome analysis of neuroblastoma cell line SH-SY5Y by GeLC-MS/MS and 2-DE.

Department of Biomedical Sciences, University of Antwerp, Antwerp Belgium.
Each eukaryotic cell in a given organism contains the same genome but the protein content of cells varies widely, depending
on cell-type, developmental stage, environmental conditions etc. Proteome analysis is usually performed by the separation of
a protein mixture by two dimensional (2DE) gel electrophoresis and the identification of the separated proteins by mass
spectrometry. Next to the fact that 2DE suffers from being labor intensive and that only high abundant proteins can be
visualized, the detection of basic, hydrophobic and low molecular weight proteins is inefficient and the complex protein
mixture exceeds the resolving power of 2DE. A simpler method for the proteome analysis is separation of the protein mixture ...

SUMMARY
The results of this investigation showed a retardation of HSV-2
replication in neuroblastoma (NB) cells without affecting HSV-1
replication. HSV-1 and HSV-2 were grown in Chang's liver (CH), human
embryonic lung (HEL), Vero and NB cells with same virus stocks. The
harvests of the infected cells were assayed for infectivity titer in Vero
cells. It was observed that the infectivity titer of HSV-2 exclusively in NB
cells was three to four logs lower in comparison to CH, HEL and Vero
cells. The clinical isolates of HSV-2 showed lower infectivity in NB cells
in comparison to CH and HEL cells. The kinetics of replication of HSV-2
in NB cells showed retardation in comparison to HSV-1. This may ...

NEUROBLASTOMA SCREENING STUDY ENABLES ADEQUATE DECISION ON FURTHER
DEVELOPMENT OF LEGISLATIVE EARLY DETECTION PROGRAMME
B. Metzinger, IKK-Bundesverband (Bergisch Gladbach)
Studies about the effectiveness of screening are rare and far between in Germany. Realization of such studies
usually is declined because of alleged ethical reasons. Consequently screening examinations are executed
without previous effectiveness checks and quality management. Especially examinations in healthy individuals
require evidence for their benefit. Necessarily the risk (including the risk of further examination) vs. benefit
(better prognosis for early detected diseases) have to be weighed.
The pilot project " Neuroblastoma screening" is a positive example for the intention of all persons concerned
realizing a study to proove the benefit of a screening test. The result of the project answers the question whether
screening ...

Introduction
Robotic integration of cell-based assays for in vitro toxicity
screening of chemical compounds is becoming an
increasingly important aspect of the drug discovery
process. Development of assays that simplify the
automation process is crucial for the success of this high-
throughput screening. Promega has recently introduced

INTRODUCTION
Neuroblastoma is a biologically very special neoplasm that is unique to child-
hood. As can be seen from Table 1 it is the fourth most common paediatric
malignancy in the Nordic countries and constitutes around 6% of all
malignancies in children.

Abstract. Valproic acid (VPA) as a differentiation inducing
anti-neoplastic substance is currently tested in solid tumour

that the anti-cancer activity of VPA was synergistically

Abstract. The majority of high-risk neuroblastomas lack the
expression of caspase-8 due to gene silencing which suggest a
mechanism for the selection of tumour cells that are refractory
to multiple cytotoxic drugs including tumour necrosis factor-
related apoptosis-inducing ligand (TRAIL). Inhibitors of DNA
methyltransferases and IFN-
Á
induce expression of caspase-8,
and sensitise some neuroblastoma cells to TRAIL-mediated
apoptosis. Here we demonstrate that a combination of cyto-
static drugs with IFN-
Á
and TRAIL synergistically induces
neuroblastoma cell death, which may have implications for
future therapy of children with neuroblastoma . Treatment of
neuroblastoma cells with IFN-
Á
induced caspase-8 expression
in all cell lines investigated. In five of the neuroblastoma cell
lines (SHEP-1, SK-N-AS, SK-N-FI, SH-SY-5Y and Kelly),

Abstract. Neuroblastoma , a form of neuroblastic tumor, is
one of the most common neoplasms seen in early childhood.
The diverse clinical behavior of this tumor is most probably
explainable by the heterogeneity in the associated genetic
changes. We investigated 12 neuroblastoma patients for
MYCN amplification and chromosome 2 aneusomy by
fluorescence in situ hybridization (FISH) and results were
correlated with conventional cytogenetics. Samples from
both primary tumor tissue and bone marrow metastasis were
available in two cases. The copy number of MYCN oncogene
paralleled that of chromosome 2 in 10 cases, whereas two
cases (16.7%) showed numerous distinct signals within the
nuclei of the tumor cells, consistent with MYCN amplification
as double minute (dmin). The morphology of dmin in one
case was of an extremely small type and might potentially be
missed by conventional chromosome analysis. ...

Abstract
Exposure to the heavy metal, cadmium (Cd2+), has been linked to adverse environmental
and human health effects. The present studies addressed the mechanisms of Cd2+-induced
cell death in the mouse neuroblastoma X rat mesencephalon (MES-23.5) wild type cell line.
Toxicity initially was indexed using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenytetrazolium
bromide (MTT) assay following exposure to Cd2+ (1-10_M) for 7 to 72 h. Cell death was
observed to be both concentration- and time-dependent. The highest concentration resulted
in 100% cell death following 72 h exposure via the culture medium. We directly evaluated ...

Dear Lunch for Life Sponsor,
The Children’s Neuroblastoma Cancer Foundation would like to give you the opportunity to be
one of our sponsors for the 2004 Lunch for Life Campaign. This campaign is set to launch on
September 1, 2004, and although the campaign runs throughout the year, we are planning special
events throughout the Pediatric Cancer Awareness Month of September.
What is Lunch for Life?
Lunch for Life gives kids an opportunity to help other kids just like themselves. These are kids
who love to play outside, swim, fly kites, and play games, but because their young lives have
been turned upside down by the words "You have cancer," they no longer can enjoy the activities
of a "normal" child. Their lives are filled with doctor visits, injections, chemotherapy,
radiotherapy, long days of hospitalization and countless hours of pain and, unfortunately,
suffering. Until a cure is found, this scene will continue ...
