Detection of Amplification in Neuroblastoma Using Polymerase Chain Reaction and Its Impact on Survival A P RATAP , S S INHA , DK G UPTA Molecular biology has revolutionized the medicine and added a new dimension to our knowledge of disease processes, especially in the field of oncology. Proto oncogenes are proteins normally present in the genome and they may be converted to oncogenes by mutations, rearrangements, translocation and deletion. One such protooncogene is the N-myc proto-oncogene seen in neuroblastoma . Amplification of the N-myc oncogene identifies a group of patients who have a poor prognosis. At present, it is generally agreed that the amplification of N-myc is still the most important prognostic factor to be investigated in every patient with neuroblastoma because of it’s reliability as a prognostic indicator. There are many conventional ...

I80 EPIDEMIOLOGY IN THE GERMAN NEUROBLASTOMA SCREENING PROJECT C. Spix, Dt. Kinderkrebsregister, Joh.Gutenberg Universität (Mainz) In Germany Neuroblastoma is the most frequent extracranial solid childhood tumor. Its properties made it seem an ideal candidate for screening. The German childhood cancer registry was involved in the project as epidemiological consultant and to ensure ascertainment of cases and deaths. A complete registry, which determines the incidence, age at diagnosis, stage distribution, and mortality in all relevant groups (control area and study area, participants and non participants) independently of the study and beyond the field phase of the study is an optimal basis for the evaluation of a screening study. Ascertainment was achieved by regular stochastic record linkage of encrypted personal identifier data from all 1.5 million study participants with the registry data at the ...

Demonstration of a significant advantage of purged ABMT compared to chemotherapy in a randomized trial, as well as improvement in survival for stage 4 >1 year of age. Implementation of novel biological therapies with demonstration of a significant benefit of 13-cis-retinoic acid for minimal residual disease in a randomized trial       Neuroblastoma Risk Groups  Survival by Stage, Age, Ferritin, Pathology, MYCN       CCG-3891:  EFS ABMT vs Chemotherapy  (as randomized)   P=0.045   ...

39 Budget: Vol. I, page 155       Overall Aims   Perform tests required for CCG therapeutic studies. Perform and facilitate laboratory - clinical correlative research dealing with biology, risk assessment, and therapy.       Research Accomplishments   MYCN and histopathology Incorporation into clinical management Confirmation of prognostic importance Identification of new subgroups Immunocytology Confirm efficacy of ex vivo purging of marrow Early identification of poor responders Biology, risk assessment, therapy Neurotrophins and receptors Molecular genetics: CGH, LOH Telomerase Metastases Cell lines:  models for developing new therapy       Required Tests   Hypothesis 1. Risk factor analysis at diagnosis will define relatively homogenous groups for selecting therapy and for evaluating ...
