Neuroblastoma Incidence in Illinois: 1986-1994
Neuroblastomas are childhood tumors of the autonomic nervous system. The annual incidence rate
among children under 15 years of age in the United States is nine to 11 cases per million. In Illinois,
according to data reported to the Illinois State Cancer Registry for 1986 through 1994, the annual incidence
rate was 10.3 per million. Table 1 lists numbers of neuroblastoma incidence among children less than 15
years of age by county in Illinois from 1986 though 1994. As one would expect, counties with a large
population, such as Cook and Du Page, tend to have more cases.
In order to detect unusually high incidence of neuroblastoma , numbers in Table 1 need to be related
to their population denominators, compared with the expected number of cases based on the overall
neuroblastoma incidence in Illinois, and expressed in terms of probabilities. This was done by calculating ...

ABSTRACT
Olfactory neuroblastoma (ONB) is a rare tumor.
There is greater agreement for separating “neuroblasto-
ma-like” forms from tumors arising in the same location
and manifest morphological and immunohistochemical
characteristics of neuroendocrine carcinoma.
The aim of present study is to describe the typical
clinical, histological and immunohistochemical findings
of ONB in 58-year old woman which help us to exclude
non-Hodgkin’s lymphoma and endocrine carcinoma.

oma like forms, neuroendocrine carcinoma

Overexpression of a DEAD Box Protein (DDX1) in Neuroblastoma

(Received for publication, November 17, 1997, and in revised form, June 2, 1998)
Roseline Godbout‡, Mary Packer, and Wenjun Bie
From the Department of Oncology, Cross Cancer Institute and University of Alberta, 11560 University Ave., Edmonton,
Alberta T6G1Z2, Canada
The DEAD box gene, DDX1 , is a putative RNA helicase
that is co-amplified with MYCN in a subset of retinoblas-
toma (RB) and neuroblastoma (NB) tumors and cell
lines. Although gene amplification usually involves hun-
dreds to thousands of kilobase pairs of DNA, a number of
studies suggest that co-amplified genes are only overex-
pressed if they provide a selective advantage to the cells
in which they are amplified. Here, we further character-
ize DDX1 by identifying its putative transcription and
translation ...

Abstract. Neuroblastoma , originated from neural crest cells, is
the most common extracranial solid tumor in childhood. In the
present study, we evaluated in vitro the oncolytic effect of live-
attenuated poliovirus on human neuroblastoma cell lines, and
in vivo its therapeutic efficacy in human neuroblastoma -
bearing athymic mice. Live-attenuated poliovirus killed 27
(93%) of 29 established neuroblastoma cell lines in vitro . It
induced cleavage of eukaryotic translation initiation factor 4G,
leading to cell death through a mechanism involving activation
of caspase-9, caspase-3 and poly(ADP-ribose)polymerase. For
the in vivo experiments, an animal model was established
using athymic mice xenotransplanted with SJ-N-JF
neuroblastoma cells on both flanks. Inoculation of live-
attenuated poliovirus into one of the two tumors caused a
dramatic and complete regression of both the inoculated and ...

My research concerns neuroblastoma ( NB ), a childhood tumor that exhibits two different phenotypes:
favorable and unfavorable. Favorable NB undergo regression or maturation even without treatment or
can be cured by surgical removal with or without adjuvant chemotherapy. In contrast, the prognosis of
unfavorable NB is poor despite the most intensive treatment. Current survival rate of children with
unfavorable NB is about 20-25%. Neuroblastoma is thus one of the last pediatric tumors to be
conquered. For decades, NB research has focused mainly on “unfavorable features” of the tumor such
as 1p deletion and MYCN amplification, assuming that these features determine the malignant
phenotype of NB. However, our studies have revealed that expression levels of a set of genes
( Favorable Neuroblastoma Genes ) in fact determine the benign or malignant phenotype of this tumor.
Favorable NB genes ...

Summary / Zusammenfassung
Common tumors in children include leukemia, brain tumors and neuroblastoma . Current
treatments of childhood malignancies such as radiotherapy and chemotherapy are inefficient, due
to the resistance of the tumor cells to apoptotic signals. Promising new therapies are, however,
emerging, which are based on blocking receptor tyrosine kinase (RTK) signalling to some of their
downstream signalling targets such as phosphoinositide 3-kinase (PI3K), protein kinase B (PKB)
/Akt, the mammalian target of rapamycin (mTOR) or mitogen-activated extracellular signal-
regulated kinase activating kinase (MEK).
We want to further study the potential of targeting PI3K signalling as a novel ...

Abstract. Thalidomide has previously been shown to have anti-
angiogenic properties. More recently, clinical efficacy of this
agent has been demonstrated in multiple myeloma and
prostate cancer. Neuroblastoma is the most frequent solid
tumor of the abdomen of childhood, yet children with this
disease frequently have metastases at presentation. Such
patients have a very poor prognosis with current therapies.
Thus, new approaches are needed. We have previously shown
that VEGF antagonists can inhibit neoangiogenesis and tumor
growth in experimental neuroblastoma . In this study, we
investigated the anti-angiogenic and anti-tumor properties of
thalidomide in a xenograft model of human neuroblastoma .
Tumors were induced in athymic mice using the human
neuroblastoma cell line NGP. Intraperitoneal thalidomide
(100 mg/kg/dose) or vehicle was administered beginning one
week after implantation, and animals ...

Neuroblastoma since its description in the late 19
th
and early 20
th
century has been well known as an enigmatic disease. Cushing and
Wolbach described in 1927 the case of a 2-year old boy whose thoracic
paravertebral sympathetic neuroblastoma over the course of 10 years
transformed into a completely differentiated ganglioneuroma.
1
The
only treatment at the time was injection of Coley’s bacterial toxins. At
the opposite end of the spectrum, the majority of neuroblastomas in
older children present with widespread metastases and have a poor
survival even with intensive chemotherapy and bone marrow trans-
plantation (BMT). Our understanding of the basis for the diverse
behavior of neuroblastoma has been greatly increased by our discov-
