Gene Therapy of Pancreatic Cancer With Green Fluorescent Protein and Tumor Necrosis Factor–Related Apoptosis-Inducing Ligand Fusion Gene Expression Driven by a Human Telomerase Reverse Transcriptase Promoter Matthew H. Katz, MD, Daniel E. Spivack, MD, Shinako Takimoto, BS, Bingliang Fang, MD, PhD, Douglas W. Burton, MS, A. R. Moossa, MD, Robert M. Hoffman, PhD, and Michael Bouvet, MD Background: Tumor necrosis factor–related apoptosis-inducing ligand (TRAIL) induces apo- ptosis in malignant cells but not in normal cells. Ad/g-TRAIL, an adenoviral vector in which expression of green fluorescent protein (GFP) and TRAIL is driven by a human telomerase reverse transcriptase promoter, has shown promise as a targeted antitumor agent. Methods: To investigate the effects of TRAIL gene therapy on pancreatic cancer , BxPC-3, MIA-PaCa-2, Panc-1, and ASPC-1 cells were treated with Ad/g-TRAIL. Transfection ...

Johnson University Surgical Unit and Department of Dietetics*, Southampton General Hospital, UK We have investigated long term survivors after pancreatectomy to determine whether specific dietary assessment and advice can improve gastrointestinal symptoms, bowel function and quality of life. Ten patients (7 men, median (range) age 61 (50–75) yr) were studied 16 (6–46) months after resection for malignancy, and after exclusion of recurrence by computed tomography. Detailed symptom enquiry, quality of life assessment (QoL) and dietary history were performed. All patients completed a 1 week weighed food dietary intake survey. Detailed advice was offered and patients were reassessed 8 weeks ...

This is the 4th leading cause of cancer deaths in the U.S. The pancreas is an organ in the upper mid-abdomen that produces intestinal enzymes to help digest food and insulin to control blood sugar. Pancreas cancer affects adults ages 35-70 and men more often than women. FREQUENT SIGNS AND SYMPTOMS • Rapid, unexplained weight loss. • Pain in the back or upper abdomen that is often relieved by bending forward. • Blood clots in veins anywhere, especially the arms and legs. This is often an early sign. • Jaundice (yellow skin and eyes) from blockage of the nearby bile duct. Jaundice is usually accompanied by intense itching. • Depression. CAUSES Unknown.   pancreatic  cancer  PANCREATIC CANCER Dr. Smoot 1/30/04   RISK FACTORS FOR PANCREATIC CANCER -The adenocarcinomas are the most common and there are also cystic and hormonal tumors.  The adenocarcinoma is the most frequent cancer found in the pancreas.  There is a hereditary chronic pancreatitis and these people are at risk for cancer in the pancreas.  It is good to screen these individuals for colon cancer as well.   MEN Type I, HPNCC and FAP patients are also at risk for pancreatic cancer .  Alcoholic pancreatitis complications may be diabetes followed by pancreatic cancer . DUCTAL ADENOCARCINOMA - Pancreatic cancer is associated with cigarette smoking, eating a lot of meat and fish (may not be a strong association with fish).  Fruits and vegetables are protective but coffee is more than likely not. -2/3 of the carcinomas occur in the head ...

PDF [35,7 KB]  From [www.berkshire.nhs.uk]  Last viewed: 30.07.2004
Berkshire Priorities Committee Policy Statement 40 Gemcitabine for Pancreatic Cancer Date of Issue August 2001 Review Date April 2004 Pancreatic adenocarcinoma causes about 11 per 100,000 deaths each year in England and Wales (just under 6,000 people). In Berkshire this equates to approximately 90 residents, national data suggests that at least 80% (70) of these residents have locally advanced or metastatic disease. It is assumed 4% would be suitable for potentially curative surgery, 25 – 35% (18 – 24) could be suitable for chemotherapy with about 50% (9-12) accepting treatment with Gemcitabine. The National Institute of Clinical Excellence (NICE) reviewed the effectiveness of Gemcitabine in five RCT studies. One single blind RCT compared Gemcitabine with the chemotherapy agent 5FU in 126 patients. The Gemcitabine patients had an 18% one year survival ...

Tel Aviv, Israel Summary Pancreatic cancer is the 10 th most common malignancy and the 4 th largest cancer killer in adults. Surgery offers the only chance of curing these patients. Complete surgical resection is associated with a 5-year survival rate of between 20 and 30%. The challenge is how to best select those patients for curative surgery. Early studies demonstrated excellent sensitivity of EUS in detecting pancreatic tumors in comparison to CT. Similarly, EUS showed an 85-94 % accuracy rate for ...

Abstract. The gastrointestinal peptides gastrin and cholecysto- kinin (CCK) stimulate growth of human pancreatic cancer by a receptor-mediated process. The purpose of this study was to investigate the molecular and functional characteristics of the receptor associated with peptide-induced pancreatic cancer proliferation. Utilizing total RNA from human pancreatic cancer cells a cDNA was cloned and sequenced by RT-PCR and rapid amplification of cDNA ends methodology. The molecular characteristics of the receptor cDNA were studied by Northern analysis and protein structure by Western analysis. An antibody raised against the novel receptor was utilized to investigate the role of the CCK-C receptor on pancreatic cancer cellular growth using in vitro technology. A spliced variant of the CCK-B receptor was identified which differed from the CCK-B receptor by the presence ...

Tangrea, Matti Rautalahti, Jacob Sehlub, Jarmo Virtamo, Philip R. Taylor Background: Few risk factors for pan- creatic cancer have been identified, with age and cigarette smoking being the most consistent. The protective ef- fect associated with consumption of fruits and vegetables—the major di- etary sources of folate—is suggestive of a role for factors influencing cellular methylation reactions; however, to our knowledge, no study has investigated this relationship. Whether biochemical indicators of methyl-group availability are associated with exocrine pancreatic cancer risk was the focus of ...
