Pancreatic cancer remains a cancer with poor survival prospects. For males
there has been no change in incidence or mortality over the last 20 years, and for
females there has been only a slight increase in both measures.

Survival regulation in pancreatic cancer cells by c-Jun
[335,4 KB]
From [147.52.72.117] Last viewed: 25.07.2004
Abstract
. Over 90% of human pancreatic cancers harbor an
activating point mutation in the K-ras gene at codon 12.
However, it is not clear whether all downstream K-ras are
activated and which downstream contributes to the cell
survival and proliferation of pancreatic cancer cells. MEK
kinase 1 (MEKK1)-c-Jun N-terminal kinase (JNK)-c-Jun
pathway has an important role in cell proliferation, survival
and apoptosis in various cells. We previously demonstrated
that the dominant negative form of MEKK1 (DN-MEKK)
inhibits the survival of human pancreatic cancer cell lines. In
this study we investigated whether JNK-c-Jun, the down-
stream pathway of DN-MEKK, affects the survival of human
pancreatic cancer cell lines. Colony formation assays
indicated that c-Jun failed to inhibit the survival of pancreatic
cancer cells, whereas c-Jun remarkably inhibited the cell
survival ...

USA (female) ? 6.2/100000

Singapore (male) ? 4/100000
Singapore (female) ? 2.8/100000
Compared to other cancers it is an uncommon cancer in Singapore.
Epidemiology
Uncommon below 45 years. 80% of cases occur in the 60-80 age group.
Male to female ration is 1.5:1 to 2:1
It remains uncertain whether geographic and racial differences in incidence reflect environmental or genetic factors. Most likely it is a combination.

Pancreatic cancer is a dismal disease.
Two years ago, 32,300 people in the U.S.
were diagnosed with pancreatic cancer
and 30,000 died. Fewer than 4 percent
of patients diagnosed survive long-term.
It is the fourth most common cause of
cancer death in the United States. In 2001,
a Progress Review Group (PRG) submitted
a detailed analysis of the state of the
disease to the National Cancer Institute
(NCI). It was the feeling of this PRG that
there had been little real
progress made in
pancreatic cancer
understanding or
treatment and that an
increased awareness
and funding on the part
of the NCI was
necessary if any real
improvements were to
be made.

Background: Evidence from both animal and human studies
suggests that abnormal glucose metabolism plays an impor-
tant role in pancreatic carcinogenesis. We investigated
whether diets high in foods that increase postprandial glu-
cose levels are associated with an increased risk of pancre-
atic cancer . Methods: In a cohort of U.S. women (n = 88 802)
participating in the Nurses’ Health Study, 180 case subjects
with pancreatic cancer were diagnosed during 18 years of
follow-up. We used frequency of intake of individual foods as

The process of pancreatic ductal adenocarcinoma development is still discussed
controversial. A key question is whether the cells of origin of the tubular complexes that
constitute precursor lesions are derived from a single cell type or from multiple types.
Suggestions that they arise solely from centroacinar cells or ductal cells have been
based on inference due to their morphological appearance in tissue from patients or
investigation of limited numbers of tubular complexes in animal models later in the
carcinogenic process. So this study was designed to monitor the development of
pancreatic cancer immediately after implantation of the carcinogen DMBA. ...

Causes of Pancreatic Cancer
Cancer of the pancreas occurs due to the mutation of DNA in pancreatic cells.
These mutations can be inherited or acquired
The inherited nature of some mutations means that pancreatic cancer seems to run in some families
Otherwise cancer of the pancreas is caused in similar ways to other cancers by exposure to carcinogens such as those in cigarette smoke.
The abnormalities in DNA are currently been investigated by techniques such as Spectral Karyotyping

UNRESECTABLE PANCREATIC CANCER

Division of General and Transplant Surgery, University of Pisa, Pisa, Italy
Background: Unbearable pain (P) in patients (pts) with unresectable or locally recurrent
pancreatic cancer (PC) is a still undertreated condition. Thoracoscopic splanchnicectomy (TS),
has recently been introduced as an attractive alternative in the management of upper abdominal P
syndromes.
Aims: The aim of this prospective study was to evaluate the effects of unilateral thoracoscopic
trunkal splanchnicectomy on P scores and quality of life measures in patients with unresectable
PC.
Materials and methods: Between September 1997 and January 2000, 36 pts (19 men and 17
women; mean age of 62, 7 years, range 48 to 82 years), underwent 38 attempted TS. Indications

4th leading cause of cancer deaths in G and E . 25,000 cases/yr in the U.S. and nearly that # of deaths. Vast majority of pts present w/unresectable disease & 5 yr survival is ~ 5%.
Cause of tumor is unknown. Risk factors include smoking & chronic pancreatitis.
1/2 of pancreatic Ca arise in the head, 1/2 in the body and tail.
Distal common bile duct runs through pancreatic head. Pancreatic Ca most often occur in this area making this tumor the most commonly associated w/malignant biliary obstruction.
Courvoisier' s law: Obstructing malignancies of the distal CBD produce a palpable non-tender gallbladder. Why? Because common duct stones (the main DDx) are usually associated w/chronic cholecystitis à contracted gallbladder.

-catenin is a component of the E-cadherin–catenin cell adhesion complex. It plays also
a role in the intracellular signalling and can function as an oncogene when it binds to the
T cell factor 4 (Tcf4)-binding site in the promoter region of cyclin D1 and transactivates
genes after translocation to the nucleus.
We evaluated the expression of -catenin in relationship with cyclin D1 overexpression,
tumor grade, clinicopathological parameters and patients’ survival in 43 adenocarcinomas

Background: We postulated an immune-mediated pathogenesis for chronic pancreatitis.
Among autoimmune pancreatitis, MFP is an inflammatory process that involves only

Introduction: Solitary metastatic lesion occurring in the pancreas is uncommon and
may be difficult to differentiate from pancreatic adenocarcinoma. Resection of metastatic
tumors to the pancreas has been occasionally reported and its role in improving survival
or quality of life of the patients is not clearly defined. We evaluated the outcome
of patients resected for isolated metastatic tumors of the pancreas, in our Institution over
a 20-years period.
Methods: From 1980 to 2000, among 222 patients resected for cancer , 6 patients who
underwent pancreatic resection for metastatic malignancies were identified. Medical

Background. Realistic models of pancreatic cancer
are necessary to develop effective drugs for the dis-
ease. More aggressive tumor models enhanced by
brighter fluorescent biomarkers to follow the disease
in real time would enhance the ability to predict ac-
curately the effect of novel therapeutics on this par-

Background: Few risk factors for pan-
creatic cancer have been identified,
with age and cigarette smoking being
the most consistent. The protective ef-
fect associated with consumption of
fruits and vegetables—the major di-
etary sources of folate—is suggestive of
a role for factors influencing cellular
methylation reactions; however, to our
knowledge, no study has investigated
this relationship. Whether biochemical
indicators of methyl-group availability
are associated with exocrine pancreatic
