[1177] Survival regulation in pancreatic cancer cells by c-Jun
PDF [335,4 KB]  From [147.52.72.117]  Last viewed: 25.07.2004
Abstract . Over 90% of human pancreatic cancers harbor an activating point mutation in the K-ras gene at codon 12. However, it is not clear whether all downstream K-ras are activated and which downstream contributes to the cell survival and proliferation of pancreatic cancer cells. MEK kinase 1 (MEKK1)-c-Jun N-terminal kinase (JNK)-c-Jun pathway has an important role in cell proliferation, survival and apoptosis in various cells. We previously demonstrated that the dominant negative form of MEKK1 (DN-MEKK) inhibits the survival of human pancreatic cancer cell lines. In this study we investigated whether JNK-c-Jun, the down- stream pathway of DN-MEKK, affects the survival of human pancreatic cancer cell lines. Colony formation assays indicated that c-Jun failed to inhibit the survival of pancreatic cancer cells, whereas c-Jun remarkably inhibited the cell survival ...

By Russell G. Postier, M.D. John A. Schilling Professor and Chairman, Department of Surgery Pancreatic cancer is a dismal disease. Two years ago, 32,300 people in the U.S. were diagnosed with pancreatic cancer and 30,000 died. Fewer than 4 percent of patients diagnosed survive long-term. It is the fourth most common cause of cancer death in the United States. In 2001, a Progress Review Group (PRG) submitted a detailed analysis of the state of the disease to the National Cancer Institute (NCI). It was the feeling of this PRG that there had been little real progress made in pancreatic cancer understanding or treatment and that an increased awareness and funding on the part of the NCI was necessary if any real improvements were to be made. At OU Physicians, we are building a substantial clinical program for ...

Abstract. Peroxisome proliferator-activated receptor Á (PPAR Á ), a ligand activated transcription factor, forms a heterodimer with retinoid X receptor · (RXR · ), and its trans- criptional activity is thought to be maximal in the presence of both PPAR Á and RXR · ligands. Though previous studies suggested that thiazolidinediones (TZDs), known as PPAR Á ligands, inhibit the growth of certain types of cancer cells, little is known about the growth inhibitory effects mediated though activation of PPAR Á /RXR · . We examined the effects of troglitazone (one type of TZDs) and 9-cis retinoic acid, a RXR · ligand, on activation of PPAR Á /RXR · and growth inhibition of human pancreatic cancer cell lines ...

Dietary Sugar, Glycemic Load, and Pancreatic Cancer Risk in a Prospective Study Dominique S. Michaud, Simin Liu, Edward Giovannucci, Walter C. Willett, Graham A. Colditz, Charles S. Fuchs Background: Evidence from both animal and human studies suggests that abnormal glucose metabolism plays an impor- tant role in pancreatic carcinogenesis. We investigated whether diets high in foods that increase postprandial glu- cose levels are associated with an increased risk of pancre- atic cancer . Methods: In a cohort of U.S. women (n = 88 802) participating in the Nurses’ Health Study, 180 case subjects with pancreatic cancer were diagnosed during 18 years of follow-up. We used frequency of intake of individual foods as reported on a food-frequency questionnaire in 1980 to cal- culate sucrose, fructose, and carbohydrate intakes; glycemic index ...

TRANSDIFFERENTIATION IS A KEY STEP IN PANCREATIC CANCER DEVELOPMENT . M.W. Mueller, D.E. Bockman, P. Buechler, J. Guo, F. Bergman*, M.W. Buechler, H. Friess. Department of General Surgery and Pathology*, University of Heidelberg, Germany The process of pancreatic ductal adenocarcinoma development is still discussed controversial. A key question is whether the cells of origin of the tubular complexes that constitute precursor lesions are derived from a single cell type or from multiple types. Suggestions that they arise solely from centroacinar cells or ductal cells have been based on inference due to their morphological appearance in tissue from patients or investigation of limited numbers of tubular complexes in animal models later in the carcinogenic process. So this study was designed to monitor the development of pancreatic cancer immediately after implantation of the carcinogen DMBA. ...

These mutations can be inherited or acquired The inherited nature of some mutations means that pancreatic cancer seems to run in some families Otherwise cancer of the pancreas is caused in similar ways to other cancers by exposure to carcinogens such as those in cigarette smoke. The abnormalities in DNA are currently been investigated by techniques such as Spectral Karyotyping   A Spectral Karyotype of a Pancreatic Cancer Sufferer                                                 &nb...

O8 PANCREATIC CANCER . FURTHER BASIC ASPECTS 29 Ectopic Cytoplasmic Expression of -Catenin Correlates with Cyclin D1 Overexpression in Pancreatic Cancer M. Ramadani, Q. Qiao, F. Gansauge, S. Gansauge, G. Leder and H.G. Beger Department of General Surgery, University of Ulm, Germany -catenin is a component of the E-cadherin–catenin cell adhesion complex. It plays also a role in the intracellular signalling and can function as an oncogene when it binds to the T cell factor 4 (Tcf4)-binding site in the promoter region of cyclin D1 and transactivates genes after translocation to the nucleus. We evaluated the expression of -catenin in relationship with cyclin D1 overexpression, tumor grade, clinicopathological parameters and patients’ survival in 43 adenocarcinomas of the pancreas and in 5 normal pancreatic tissues by immunohistochemistry.
