Background: Few risk factors for pan-

with age and cigarette smoking being
the most consistent. The protective ef-
fect associated with consumption of
fruits and vegetables—the major di-

BACKGROUND: Interleukin (IL)-13 and -4 are multifunctional cytokines that
bind to specific cell-surface receptors. The aim of this study was to determine
whether pancreatic cancer cells express either receptor, and to assess the growth
suppressive effects of chimeric proteins composed of a Pseudomonas exotoxin
(PE) A mutant (PE38QQR) fused to IL-13 (IL-13-PE38QQR) or IL-4
(IL-4-PE38QQR) in these cells. MATERIALS AND METHODS: Northern and
Western blot analysis were used to analyze the expression of IL-4/-13 receptors
and the common gamma chain (gamma c) in pancreatic cancer cell lines. MTT

Pancreatic cancer cells are usually resistant to apoptosis induced by cytotoxic
drugs, by activation of surface receptors such as Fas and TNF receptor or by
serum or growth factor withdrawal. Actinomycin D (actD) is an inhibitor of
RNA synthesis and acts as a potent inducer of apoptosis in several cell lines.
In the present study, we investigated the effects of actD on PANC-1
pancreatic cancer cells. ActD caused apoptosis in PANC-1 cells in a
dose-dependent manner, as determined by cell growth assays, DNA laddering
and TUNEL assays. Induction of apoptosis correlated with activation of the

Department of Medicine, University of California, Irvine, USA.
Cyclin D 1 belongs to a family of protein kinases that have been implicated in
cell cycle regulation. In the present study we characterized cyclin D1
expression in 6 cultured human pancreatic cancer cell lines and in normal and
cancerous human pancreatic tissues. A 4.4-kb cyclin D1 mRNA transcript was
present in all cell lines and in all pancreatic tissues. Cyclin D1 mRNA levels
were 2.1-fold higher in the pancreatic cancers than in normal pancreatic tissues
(p < 0.0002). Cancer patients with lower cyclin D1 levels (n=16) had a median
survival of 15.5 months whereas patients with higher levels (n=16) had a
median survival of 6.5 ...

Department of Medicine, University of California, Irvine 92697, USA.
OBJECTIVE: To investigate Fas and FasL expression in pancreatic tissues and
cultured pancreatic cancer cell lines, and to assess the ability of anti-Fas
antibodies to induce apoptosis. SUMMARY BACKGROUND DATA:
Activation of the Fas receptor by Fas-ligand (FasL) results in apoptosis, and
dysregulation of this pathway may contribute to abnormal cell proliferation.
METHODS: Northern blotting and immunohistochemistry were used to
compare Fas and FasL expression in normal and cancerous tissues. DNA 3'-OH
end labeling was used to detect apoptotic cells. The effects of Fas activation on
cell growth and signaling pathways were investigated in culture. RESULTS:
Pancreatic cancers exhibited increased Fas RNA levels, whereas ...

Dear Sir:
We have read the informative article about
Helicobacter pylori (HP) and pancreatic
disease written by Manes et al . with great
interest [1]. We were, however, slightly
puzzled by the statement that no data on a
potential association between Helicobacter
pylori infection and pancreatic cancer are
available. According to recent data, two
articles and one letter about an association
between the bacteria and pancreatic cancer
have been published. In the ...

Pancreatic Cancer and
Exposure to Ionizing Radiation
Summary
: Some evidence has been recorded of a possible connection between cancers of the
pancreas and exposure to ionizing radiation. This possible connection is supported by evidence
from studies conducted at Los Alamos National Laboratory and other studies of nuclear workers
at other sites who have been exposed to ionizing radiation. The National Research Council’s, on
the other hand, has determined that the pancreas is relatively insensitive to ionizing radiation.

Pancreatic cancer is a dismal disease.
Two years ago, 32,300 people in the U.S.
were diagnosed with pancreatic cancer
and 30,000 died. Fewer than 4 percent
of patients diagnosed survive long-term.
It is the fourth most common cause of
cancer death in the United States. In 2001,
a Progress Review Group (PRG) submitted
a detailed analysis of the state of the
disease to the National Cancer Institute
(NCI). It was the feeling of this PRG that
there had been little real
progress made in
pancreatic cancer
understanding or
treatment and that an
increased awareness
and funding on the part
of the NCI was
necessary if any real
improvements were to
be made.

HER2 signaling is associated with enhanced breast cancer cell proliferation. The aims of this study are to investigate if
HER2 in human prostate, breast and pancreatic cancer cells is a molecular target of a new treatment with targeted alpha

Introduction: The signaling pathways that regulate pancreatic cancer growth are poorly
understood. 5-lipoxygenase (5-LOX) is involved in pancreatic cancer cell proliferation.
The 5-LOX metabolite, 5-hydroxyeicosatetraenoic acid (5-HETE) stimulates pancreatic
cancer cell proliferation via activation of MAPK. The roles of Rac1/cdc42 in 5-HETE-
induced MAPK activation were investigated.
Materials and Methods: Phosphorylation of ERK1/2, P38 kinase, PAK and Raf-1 were
measured by western blotting. Activities of Rac1 and cdc42 were measured by selective

Introduction: The factor and the mechanism that determine the metastatic and invasive
behavior of pancreatic tumor cells are incompletely understood.
Methods: The expression of integrin subunits and the alteration of their expression by
GDNF were examined by flow-cytometric analysis and cellular enzyme-linked assay
in pancreatic cancer cell lines. Adhesion and invasion assays of cancer cells to ECM
proteins were performed.
Results: Pancreatic cancer cell lines were all found to express the RET and GFR -1
