• Thyroid cancer is the most common endocrine cancer .
• It occurs in all age groups, mainly adults. It affects about three times
as many women as men.
• There are several types—papillary, follicular, medullary, anaplastic,
and variants.
• Thyroid cancer is one of the few cancers that is increasing in incidence—
with 17% more new cases in 2006 than in 2005. It's #1 in incidence
growth in women and #3 in men.
• About 30,180 people, including 22,590 women and 7,590 men, will be
diagnosed with thyroid cancer in 2006 in the United States, according
to the American Cancer Society. About 870 women and 630 men (1,500
total) will die of thyroid cancer in 2006.

The incidence of thyroid cancer has been rising gradually over the past decade. After initial treat-
ment with thyroidectomy, usually followed by adjuvant radioiodine ablation and thyroid hormone
suppressive therapy, most patients will be cured and remain cancer free. However, persistent or recur-
rent disease is not uncommon. Drs. Tuttle and Mazzaferri discuss the rapidly evolving approaches to
detect thyroid cancer recurrence, and the therapeutic challenges posed by the increased sensitivity
of these new surveillance modalities. Current therapies are ineffective for those patients who have
metastatic cancer unresponsive to surgery and radioiodine. Dr. Fagin discusses how the knowledge of
the genetic origin of thyroid cancer is being used to design and test promising new therapies.

Thyroid cancer is the most common endocrine-related cancer ;
however, it is rare compared to other cancers. In the United States
there are only about 20,000 new patients annually. Even though the
diagnosis of cancer is terrifying, the outlook for patients with thyroid
cancer is usually excellent. First, most thyroid cancer is easily curable
with surgery. Second, thyroid cancer rarely causes pain or disability.
Third, effective and well-tolerated treatment is available for the most
common forms of thyroid cancer .

Papillary/Follicular Cancer :
Papillary and follicular cancer , or carcinoma, account for about 80-85% of all
thyroid malignancies. They get their names from their appearance under the
microscope. Some are pure papillary, some are purely follicular in nature, and
some are mixed, that is, both types of cancer cells may be present in the
same tumor. In fact, some of these tumors are actually referred to as
“follicular variant of papillary carcinoma”. The important thing to understand
is that 1) the typical forms of these cancers are quite curable in a very large
percentage of cases and, 2) they behave in somewhat dissimilar ways, and
affect slightly different age groups, though there are often exceptions to the
rule.

7-Hydroxystaurosporine (UCN-01) is a selective protein kinase C (PKC) inhibitor and is being developed as a
novel anticancer agent. Because of reports that PKC may be involved in the pathogenesis of some forms of thy-
roid cancers, we examined four thyroid carcinoma lines (FRO, KAT5, NPA, and WRO). These cells were found
to have different susceptibility to UCN-01 treatment, and there appeared to be a correlation between UCN-
01–induced death and expression levels of endogenous Bcl-2. KAT5 cells, which normally express a low amount

Cancer of the thyroid made up about one percent (625 cases) of the invasive
cancers diagnosed in Ohio in 1999 and reported to the Ohio Cancer Incidence
Surveillance System (OCISS) (Table 1). For 1999, the Ohio age-adjusted
thyroid cancer incidence rate was 5.5 cases per 100,000 residents, which is 24

iny and usually insignificant carcinomas
can be found in five to 10% or more of all thyroid
glands that are carefully examined under the
microscope at autopsy, but relatively few of them
grow or spread to produce symptoms that lead to
their detection during a person’s lifetime. The thyroid
cancers that are diagnosed each year represent about 1% of
all cancers in the U.S. population. Most types of thyroid
cancer rank quite high in terms of successful
treatment and long term survival. However,
some rare subtypes may have a poor
prognosis.
The highest incidence rates for
thyroid cancer in the SEER regions occur in
